Elmalarda İmidacloprid ve Dimethoate Pestisitlerinin Kalıntılarının Belirlenmesinde QuEChERS ve LC MS MS ile Metot Validasyonu

Özet

Metot validasyonu (geçerli kılma) pestisit kalıntı analizlerinde önemli bir kalite parametresidir. Pestisit kalıntı analizlerinde kullanılan tüm metotların, orijinal laboratuvar örneklerinin analizinden önce validasyonu yapılmalıdır. Bu çalışma elmalarda imidacloprid ve dimethoate kalıntı analizi için güvenilir bir analiz prosedürünü valide etmek için gerçekleştirilmiştir. Bu amaçla hiçbir ilaçlama yapılmamış Golden Delicious ve Starking Delicious elma örnekleri iyi bir şekilde homojenize edilmiş ve pestisitlerle 3 farklı seviyede zenginleştirilmiştir. 10 g elma örneği QuEChERS yöntemi ile ekstraksiyon ve clean up (istenmeyen bileşiklerden arındırma) işlemine tabi tutulmuştur. Sonra da LC-MS/MS sisteminde imidacloprid ve dimethoate analizleri yapılmıştır. Her iki pestisit ve elma çeşiti için matris etkisi Student t testi ile önemli bulunduğundan, miktarsal hesaplama matrisli kalibrasyon (MC) ile yapılmıştır. Analiz metodunun validasyonu, geri alım, metot dedeksiyon limiti, tekrar edilebilirlik, kesinlik gibi metot performans kriterleri ile gerçekleştirilmiş ve tümü olması gereken limitler içinde olmuştur. Her iki elma çeşiti için imidacloprid ve dimethoate geri alım değeri ortalaması %88.34 (%7.72 oransal standart sapma ile) olarak bulunmuştur. Tüm metodun geri alımı ise  %12.02 oransal standart sapma ile %89.50 olarak bulunmuştur (n = 150). Bu rakamlar olması gereken ortalama geri alım (%70–120) ve tekrar edilebilirlik  (RSD ≤ 20%) değerleri ile uyumludur. Pestisitlerin metot dedeksiyon limiti,  AB ve Türk Gıda Kodeksi MRL değerlerinden düşük bulunmuştur. Her iki pestisit için kalibrasyon eğrisi 5–200 ng mL-1 konsantrasyon sınırlarında doğrusal bulunmuştur.

Method Validation for Determination of Imidacloprid and Dimethoate Pesticides in Apples by QuEChERS and LC–MS/MS Analysis.

 

 Method validation is an important quality parameter in the pesticide residue analysis. All methods used for pesticide residue analysis must be validated prior to implementation of original laboratory samples analysis. This study was carried out to validate a reliable analytical procedure for imidacloprid and dimethoate residues in apples. For this aim, untreated blank Golden Delicious and Starking Delicious apple samples were uniformly homogenized and fortified at 3 levels with pesticides. 10 g of apples were subjected to QuEChERS extraction method, incorporating a simple simultaneous cleanup step, followed by imidacloprid and dimethoate analysis with LC-MS/MS. According to Student t test matrix effect is important for both pesticides and both apple varieties. Thus, matrix matched calibration (MC) was used for quantification. The validity of analytical method was performed by the method-performance criteria such as, recovery, method detection limit, repeatability, precision, and all found to be within the required limits. For imidacloprid and dimethoate analytes, recoveries averaged 88.34% (with a relative standard deviation of 7.72 %) for two apple commodities over the validation range. For  overall recovery of the method was  89.50 % with a relative standard deviation of 12.02% (n = 150). These figures comply with the values specified for mean recovery range (70–120%) and repeatability (RSD ≤ 20%).  Estimated method detection limits of analytes were below the EU MRLs and also TGK MRLs. Calibration curve of two compound were also linear within the linear range of 5–200 ng mL-1 .
