Trehalose is required for stress tolerance and regulation of glucose uptake. Neutral trehalase enzyme, encoded by NTH1 gene, responsible from the hydrolysis of stress accumulated trehalose in Sacchaoromyces cerevisiae. Transcription of NTH1 gene is activated by Msn2/4 transcription factors which are stress activated transcription factors. In this study, we have analyzed if the chromatin modifying complexes involves in the transcription of NTH1 gene under normal and also under stress inducing growth conditions. First we have analyzed the nucleosome positioning profile on the 1 Kbp region of NTH1 promotor region using Chromatin Folding v2 software. The nearest two nucleosomes to transcription start site overlaps with the STRE elements for binding to Msn2/4p. Then we have investigated whether nucleosome mobilizing complexes is involved in the NTH1 transcription or not. We tested the effect of Spt7p, subunit of SAGA complex, on the transcriptional regulation of NTH1. We have found that transcription of NTH1 gene increased by at least 3-fold in Δspt7 mutants than wild type yeast strain, indicating that SAGA complex is essential for the basal level transcription of NTH1. In addition, we found that NTH1 transcription is 2-fold decreased in the Δspt7 mutants than wild type yeast under nutrient starvation. Also, when both mutant and wild type yeast strains growing rich nitrogen source, glutamine, NTH1 gene expression in wild type yeast strain increased 4 times but no activation was seen in Δspt7 mutants. Our results indicated that SAGA nucleosome mobilizing complex is essential for the basal level transcription of NTH1 gene expression.
